Given enough time in an open environment and a static source such as a tweeting bird, I can work out the direction. But a moving ambulance is impossible, and a sound that bounces around corridors is very difficult.

I use volume to determine location. If the sound is louder, then I know my ear is pointed where the sound is coming from. I will then rotate my head and see if the sound increases or decreases in volume. This is the only way I have been able to determine where sound is coming from.
